ABSTRACT

Abstract Transplanting time and genotype contribute to improving crop yield and quality of eggplant (Solanum melongena L.). A field experiment was conducted to investigate the impact of foliar applied of triacontanol (TRIA) and eggplant genotypes 25919, Nirala, 28389 and Pak-10927,transplanted on 1 March,15 March, and 1 April on exposure to high air temperature conditions. The experiment was performed according to Randomized Complete Block Design and the data was analyzed by using Tuckey,s test . The TRIA was applied at 10µM at flowering stage; distilled water was used as the control. Rate of photosynthesis and transpiration, stomatal conductance, water use efficiency, and effects on antioxidative enzymes (superoxide dismutase, catalase and peroxidase) were evaluated. The 10µM TRIA increased photosynthesis rate and water use efficiency and yield was improved in all genotypes transplanted at the different dates. Foliar application of 10µM TRIA increased antioxidative enzyme activities (SOD, POD & CAT) and improved physiological as well as biochemical attributes of eggplant genotypes exposed to high heat conditions. Highest activity of dismutase enzyme 5.41mg/1g FW was recorded in Nirala genotype in second transplantation. Whereas, lowest was noted in PAK-10927 (2.30mg/g FW). Maximum fruit yield was found in accession 25919 (1.725kg per plant) at 1st transplantation with Triacontanol, whereas accession PAK-10927 gave the lowest yield (0.285 kg per plant) at control treatment on 3rd transplantation. Genotype, transplanting date and application of TRIA improved growth, yield and quality attributes under of heat stress in eggplant.

ABSTRACT

Abstract Given the current climate change and high-temperature impact on human health, especially due to global warming, actions for heat prevention focused on outdoor workers are relevant. This essay presents the results of recent research and highlights technological innovations for heat monitoring in Brazil. Also, it reflects on the future of outdoor work given climate change. Outdoor work is generally characterized as heavy, leading to higher metabolic heat production. However, Occupational Safety and Health have made few efforts to find alternatives to minimize or eliminate heat impact on workers. Moreover, the worker's protection alternatives are reduced. WBGT Monitor is presented as a technological innovation for remote heat monitoring in outdoor environments. Despite the control measures provided to workers subjected to heat, global warming demands innovative interventions and special exposure controls at outdoor workplaces. Thus, an increased commitment to knowledge production to elaborate or improve public policy is desirable.

ABSTRACT

Heat stress refers to a series of stress reactions such as heat balance disturbance and physiological dysfunction when the body is exposed to the thermal environment for a long time. Studies have found that heat stress can damage intestinal morphology, such as length of intestinal villi, number of goblet cells, and depth of the crypt, affecting the digestion and absorption functions. It also can increase the permeability of the intestinal barrier by damaging the tight junction of the intestinal epithelium, which in turn allows endotoxin and bacteria to enter the blood circulation from the intestinal cavity to cause a systemic inflammatory response. At the same time, heat stress can disrupt the homeostasis of intestinal microbiota, increase pathogenic bacteria, and change downstream metabolites such as short-chain fatty acids. In addition, heat stress can inhibit the occurrence of hippocampal neurons and reduce the number of neurons; decrease the density of synapses; damage important organelles of neurons; induce inflammation of the central nervous system, and then lead to cognitive dysfunction. The brain-gut axis is a two-way signal axis between the intestine and the brain. Intestinal microorganisms and the intestinal barrier can participate in central nervous system regulation, and the brain can change the intestinal homeostatic function and affect the quality of the intestinal barrier through the hypothalamic-pituitary-adrenal axis (HPA axis). The interaction plays an essential role in the body's homeostasis. Therefore, this article reviewed current understandings on the impacts of heat stress on the gut and cognitive function, aiming to provide a reference for subsequent research.

ABSTRACT

Pinellia ternata is an important medicinal plant, and its growth and development are easily threatened by high temperature. In this study, comprehensive research on physiological, cytological and transcriptional responses to different levels of heat stress were conducted on a typical phenotype of P. ternata. First, P. ternata exhibited tolerance to the increased temperature, which was supported by normal growing leaves, as well as decreased and sustained photosynthetic parameters. Severe stress aggravated the damages, and P. ternata displayed an obvious leaf senescence phenotype, with significantly increased SOD and POD activities (46% and 213%). In addition, mesophyll cells were seriously damaged, chloroplast thylakoid was fuzzy, grana lamellae and stroma lamellae were obviously broken, and grana thylakoids were stacked, resulting in a dramatically declined photosynthetic rate (74.6%). Moreover, a total of 16 808 genes were significantly differential expressed during this process, most of which were involved in photosynthesis, transmembrane transporter activity and plastid metabolism. The number of differentially expressed transcription factors in MYB and bHLH families was the largest, indicating that these genes might participate in heat stress response in P. ternata. These findings provide insight into the response to high temperature and facilitate the standardized cultivation of P. ternata.

ABSTRACT

@#Workplaces in tropical countries are associated with increasing temperature and humidity, thus, the workers are susceptible to heat hazards. The inability for self-cooling properly toward exposure to head hazards can lead to severe dehydration and even death. To date, there are several indices and models to assess heat stress, such as WBGT, ISO 7933, discomfort index, HIS, PhSI, etc. However, their accuracy and suitability for tropical regions are still under investigation since they are mostly developed for subtropical regions. This review assessed the indices to estimate heat exposure in tropical regions based on the various online database. Among those indices, WBGT is the most suitable despite its inability to estimate human thermal response in tropical regions with several adjustments. Based on this review, it showed that there is a need for improvement of current indexes to be used in tropical regions since they are more suitable for the subtropical countries.

ABSTRACT

ABSTRACT: This study evaluated: (1) the vulnerability of broiler houses with different cooling systems, and (2) the spatial distribution of environmental variables during hot days. Four potentially vulnerable commercial broiler houses in southern Brazil were selected according to the following parameters: absence or presence of different cooling systems, broilers older than 28 days, and outside air dry-bulb temperature over 30°C. Broiler house vulnerability was classified according to the cooling and mechanical ventilation system: cellulose pad cooling (CPC), sprinkling (SPK), fogging (FOG), and mechanical ventilation without evaporative cooling system (VTL). The air dry-bulb temperature (Tdb, °C) and relative humidity (RH, %) were recorded every 10 min. For each broiler house, we evaluated: (1) relative cooling efficiency (RCE) and (2) inside spatial distribution of microclimate variables using a geostatistical technique. The CPC and SPK did not differ (P<0.05) in RCE (81.6% and 80.7%, respectively), but both differed from FOG (23.8%) and VLT (1.87%) systems. The highest variations in indoor Tdb were recorded in the FOG (7 °C), followed by the SPK (4 °C) and CPC (3 °C). In the CPC, there was an increase in RH from the middle to the end of the broiler house near the exhaust fans. In conclusion, the relative cooling efficiency and the inside spatial distributions of environmental variables in the broiler houses were influenced by the existing cooling system.

ABSTRACT

Objective:To explore the molecules mechanism of Pin1 in severe heat stroke induced acute lung injury by observing Pin1 regulate oxidative stress and apoptosis formation in pulmonary microvascular endothelial cells (PMVECs) and lung tissue in heat stressed mice. Methods:In vitro, a PMVECs heat stress (HS) model was established. In the control group, PMVECs were placed in a standard 37 °C, 5% CO 2 cell incubator; in the HS group, PMVECs were placed in a 43 °C cell incubator for 2 h, then the cells were further incubated at 37 °C for 1, 3, 6 or 12 h. PMVECs were pretreated with Pin1 inhibitor Juglone (1 μmol/L) 1 h before 43 °C of HS. In vivo, a severe heat stroke mouse model was established. In the HS group, the mice were kept at the simulation of climate chamber with temperature (35.5±0.5) °C, humidity (60±5)%, the rectum temperature in mice was measured by the anal rectal temperature table, when the temperature reached 42 °C, the heat exposure was stopped, and the mice were sacrificed at 1, 3, 6 or 12 h after HS. In the control group, the mice were kept at room temperature (25±0.5) °C. Mice received daily intraperitoneal administration of Pin1 inhibitor Juglone (1 mg/kg) for 3 d before HS. The protein level of Pin1, cleaved caspase-9 and cleaved caspase-3 were analyzed by Western blot, the level of O 2-˙ in cells was observed by DHE staining and fluorescence microscopy, the levels of malondialdehyde (MDA) and the activity of superoxide dismutase (SOD) in lung tissue were measured by ELISA, the pathological changes of mice in different group were detected by HE staining, and the expression of Pin1 in the lung tissue of different groups was detected by immunohistochemical staining, the apoptosis rate of the lung tissue in different groups was tested by TUNEL staining. Results:At 1 h after HS, the protein expression of Pin1 in PMVECs and lung tissue began to increase in a rewarming time-dependent manner ( F=771.6, P<0.05; F=1 035, P<0.05). Cleaved caspase-9 protein in PMVECs and lung tissue started to increase at 3 h post-HS, then increased with a rewarming time-dependent manner ( F=729.8, P<0.05; F=1 773, P<0.05). The protein expression of cleaved caspase-3 in PMVECs and lung tissue also started to increase at 3 h after HS and the expression continued to be increased with prolonged rewarming time, and the trend was consistent with cleaved caspase-9 ( F=1 084, P<0.05; F=1 252, P<0.05). In addition, HS induced the increased release of O 2-˙ from PMVECs, HS induced the imbalance of oxidation-antioxidant system in lung tissue of mice after HS which verified by the continuous release of MDA ( F=114.2, P<0.05) and the continuous inhibition of SOD activity ( F=99.15, P<0.05). Compared with the HS group, pretreatment with Pin1 inhibitor Juglone in PMVECs and mice before HS significantly inhibited the protein expression of Pin1, cleaved caspase-9 and cleaved caspase-3 (all P<0.05), pretreatment with Pin1 inhibitor greatly reduced the release of O 2-˙ in PMVECs after HS, and promoted the restore of the oxidation-antioxidant system balance of lung tissue in mice with severe heat stroke. In addition, compared with the HS group, inhibiting the expression of Pin1 significantly decreased HS induced MDA release [(11.53±0.84) nmol/mL vs (9.65±0.69) nmol/mL, t=12.52, P<0.05], promoted the restore of SOD activity [(41.18±3.45) U/mL vs (57.52±4.83) U/mL, t=5.57, P<0.05] and improved the pathological damage of lung tissue as well as decreased the occurrence of apoptosis in post-HS mice. Conclusion:It was confirmed that Pin1 is involved in heat stress induced acute lung injury mainly through mediating oxidative stress response and apoptosis.

ABSTRACT

Temperature is one of the major environmental signals controlling plant development, geographical distribution, and seasonal behavior. Plants perceive adverse temperatures, such as high, low, and freezing temperatures, as stressful signals that can cause physiological defects and even death. As sessile organisms, plants have evolved sophisticated mechanisms to adapt to recurring stressful environments through changing gene expression or transcriptional reprogramming. Transcriptional memory refers to the ability of primed plants to remember previously experienced stress and acquire enhanced tolerance to similar or different stresses. Epigenetic modifications mediate transcriptional memory and play a key role in adapting to adverse temperatures. Understanding the mechanisms of the formation, maintenance, and resetting of stress-induced transcriptional memory will not only enable us to understand why there is a trade-off between plant defense and growth, but also provide a theoretical basis for generating stress-tolerant crops optimized for future climate change. In this review, we summarize recent advances in dissecting the mechanisms of plant transcriptional memory in response to adverse temperatures, based mainly on studies of the model plant

ABSTRACT

Objective@#To investigate the expression of phosphoglycerate mutase 1 (PGAM1) in the mouse testis after exposure to single heat stress (SHS).@*METHODS@#We randomly assigned 32 C57 male mice to an SHS (n = 16) and a control group (n = 16), the former bathed in water at 43 ℃ and the latter at 25 ℃ for 15 minutes. At 1 and 7 days after exposure, we harvested the testicular tissue for observation of the morphological changes of testicular cells by HE staining and determination of the location and expression of the PGAM1 protein by immunohistochemistry and Western blot.@*RESULTS@#The testis volume of the mice were reduced significantly, the spermatogenic tubules were disorganized, and the cells were reduced in number after heat stress and basically disappeared after 7 days. Immunohistochemistry showed extensive expression of the PGAM1 protein in the testicular spermatogenic tubules of the SHS-exposed mice, significantly higher than in the control group at 1 day after exposure, which was down-regulated in the testis tissue at 7 days, but still markedly higher than that in the control. Western blot exhibited significantly up-regulated expression of the PGAM1 protein after heat stress compared with that in the control group.@*CONCLUSIONS@#The expression of the PGAM1 protein undergoes dynamic changes in the mouse testis after exposed to single heat stress, which is related to heat stress-induced proliferation and division of testicular spermatogenic cells.

ABSTRACT

Abstract Heat stress impact on physiological and behavioral parameters in dairy calves. Among the stress factors to which dairy cattle is exposed heat is one of the most studied due to the impact it has on milk production. However, there is lack of information regarding this stress condition in calves compared to the one related to adult animals. The aim of this paper was to study the relevance of heat stress in dairy calves. For the experiment 246 dairy calves from 19 dairy farms were monitored in days with Temperature Humidity Index (ITH) above 72 between 12:00 pm and 03:00 pm. Heat stress related to ITH was categorized in three degrees: low, moderate and sever stress. Additional information was taken during the visit: shade access, water availability, body position, activity being developed, rectal temperature (TR) and respiratory rate (FR). Data were analyzed using ANOVA. Comparison between median values was analyzed with Tukey´s test (α=0,05) and correlation analysis with Spearman´s test. The software used to analyze statistical data was Infostat version 2019. Differences were found in the body position between animals exposed to sever stress versus those with moderate stress (p=0.0138). However, there were no differences in animal activity in any of the temperature exposures. Results on average rectal temperature (TR) showed differences (p=0.0063) between animals with low stress and those with severe and moderate stress (> 39.2 °C). Relative average of FR was higher (p<0.0001) on calves under sever heat stress, reaching above 80 breaths per minute. The correlation coefficient between ITH and TR was 0.25 (p=0.0001), and between ITH and TR was 0.33 (p<0.0001). Regarding the practices implemented on the farms was noted that 26% of calves did not have shade or water available at the moment data was taken.

ABSTRACT

The objective of this work was to evaluate the genetic diversity of the HSP-70.1 gene and to associate the polymorphisms found with the performance of Holstein, Girolando (5/8H-G) and Sindi dairy cows raised in region of the Brazilian semiarid. Polymorphisms were identified and evaluated using the PCR-RFLP technique using the EcoRII restriction enzyme. Genetic variability was evaluated using the standard diversity index and molecular variance analysis (AMOVA). The identified polymorphisms were evaluated on the characteristics of milk production. They were identified from the seven alleles, demonstrating that there was polymorphism for the analyzed gene region and some alleles were shared among the herds. The Holstein and Sindi bovine breeds were genetically like the analyzed gene. AMOVA demonstrated that there is genetic variation between and within the herds, with most of the variation occurring within the herds for all groups evaluated. There was effect of the alleles identified on the production of milk herds of Holstein and (P<0.0001) Girolando (P<0.0117) breeds. The HSP-70.1 gene was polymorphic in the population of dairy cattle studied, and therefore a promising molecular marker to evaluate milk production of breeds created in semiarid regions.(AU)

ABSTRACT

Droughtmaster is a tropical breed of beef cattle that can survive in hot climates and easily adapt to torrid environments. These traits are important in livestock breeding. In this study, we genotyped five single-nucleotide polymorphisms (SNPs) of the AHSA2 gene from 190 cattle belonging to three different breeds (Droughtmaster, Angus and Simmental) by using snapshot technology. This work aimed to identify the valuable molecular marker of heat resistance in cattle. Results showed that Droughtmaster exhibited higher expected heterozygosity and polymorphic information content compared with the two other breeds. The AHSA2-1 locus deviated from the Hardy– Weinberg equilibrium in the Droughtmaster breed (P \ 0.05). Two SNPs in Droughtmaster diverged significantly from Angus and Simmental. The SNPs were identified as AHSA2-3 and AHSA2-4, which were closely linked to the three breeds based on pair-wise FST. AHSA2-4 involved a missense mutation. In summary, the GG genotypes in AHSA2-3 and AHSA2-4 may be candidate genotypes associated with heat resistance traits and may serve as valuable genetic markers for breeding of heat-tolerant beef cattle in the future.

ABSTRACT

Abstract Background: Heat stress derived from global warming is causing major economic losses in the livestock industry. Objective: To develop a novel methodological approach for determining the influence of climatic factors on the estimation of genetic parameters for growth traits in Retinta cattle breed by using reaction-norm models. Methods: Live weight records (n=7,753) from 3,162 Retinta calves born from 1,249 dams and 85 sires and raised in the Andalusian region (Spain) were analyzed. The effect of heat stress was measured using the temperature-humidity index, calculated with climatological data obtained from four weather stations. A bivariate-random-regression reaction-norm model was used to estimate the (co)variance components of weight until weaning in two different climatic seasons corresponding to warm and cold months. Results: The heritability pattern of individuals reared under diverse environments during the first 90 days of age was different. However, differences were not significant at the end of the growing period. Weaned calves reared during the cold season showed greater growth from 70 to 160 days in comparison with those reared during the warm season. Conclusions: Overall, this assessment did not show significant effects of the genotype-environment interaction. However, highly significant evidence of genotype-climatic condition interaction was found during the calf´s first three months of age.

ABSTRACT

Aim: This study was carried out to investigate the effect of dietary metabolic energy on growth performance, homeostasis of blood biochemical parameters, immunoglobulin G (IgG), and corticosterone in meat duck exposed to heat stress.Methodology: A total of 320 meat ducks (Cherry valley, Anas platyrhynchos) were randomly allotted into five groups. In all experimental treatment groups, dietary crude protein content was fixed at 18%. metabolic energy level for the control group (thermononeutral temperature, 26°C) was set at 3000 kcal kg-1, while metabolic energy levels for heat stress groups were set at 2900, 3000, 3100 or 3200 kcal kg-1. Heat stress conditions were artificially induced at 36°C with relative humidity of 70% once daily (from 11:00 to 16:00 hr) during finisher periods (day 22 to 42). Results: Compared to control group, heat stress groups with different metabolic energy showed significant decrease in body weight gain, feed intake, red blood cell, and platelets. Heat stress groups with different metabolic energies showed significant increase in feed conversion ratio, blood lipid profiles, aspartate aminotransferase, alanine transferase, electrolytes, pH, gas concentration, immunoglobulin G, and corticosterone compared to the control group.Among heat stress groups, red blood cells and platelets were significantly higher in the heat stress group with metabolic energy of 3000 compared to those in other groups. Blood IgG and corticosterone levels were significantly lower in 2900 and 3000 groups than those in heat stress groups with metabolic energy of 3100 and 3200. Interpretation: The results of this study suggest that dietary metabolic energy 3000 kcal kg-1 level can improve the growth performance of meat duck exposed to heat stress by modulating homeostasis of blood biochemical parameters.

ABSTRACT

Abstract Since the 1990s, an increase in the prevalence of chronic kidney disease (CKD) in several countries in Central America began to be noticed. This increase has occurred mainly in agricultural workers within that region, who are subjected to high temperatures, which led to its designation as Mesoamerican Nephropathy (MeN). Although its etiology is not clarified, it is considered that there is an occupational and environmental component involved. In this article, its main characteristics are described, including what is known about its possible etiology, diagnosis and prevention and treatment strategies.
